Abstract

The development of the immune system has been hypothesized to be a threepart process that discards useless cells (death by neglect), retains useful cells (positive selection), and deletes or inactivates dangerous cells (negative selection). Studies in the last few years in mice transgenic for antigen receptors have strongly indicated that this hypothesis is essentially correct (von Boehmer 1994; Nossal 1994). Although substantial information has recently been acquired about the editing process by which self-tolerance is imposed, our understanding of the mechanisms and genetic basis of pathogenic autoimmune syndromes is far from resolved.
