Zusammenfassung
Von Willebrand-Faktor (vWF) wird im menschlichen Körper von Endothelzellen und Megakaryozyten als primäres Translationsprodukt einer Größe von 2813 Aminosäuren synthetisiert. Im Laufe der intrazellulären Reifung wird von diesem Protein zunächst ein 22 Aminosäuren langes Sekretionspeptid abgespalten, und die entstehenden Monomere dimerisiert, sulfatiert und glykosyliert. Anschließend folgt die Multimerisierung der Dimere und die Abspaltung der 741 Aminosäuren langen Propeptide. Die unterschiedlich langen Multimere (bestehend aus einer variablen Anzahl der 2050 Aminosäuren langen, bzw. ca. 250 Kd großen Monomere) werden schließlich vom Endothel ins Blut sezerniert bzw. in speziellen Strukturen, den Weibel-Palade-Körpern, gespeichert. In Thrombozyten findet die vWF-Speicherung in den sog. a-Granuolen statt [10,11].
