Abstract
We analyzed the N-RAS, K-RAS, H-RAS and FLR exon of the neurofibromatosis 1 (NF1) gene in 11 patients at the time of diagnosis of a secondary acute leukemia. Eight of these patients suffered from secondary acute myelogenous leukemia (AML) and three patients had a secondary acute lymphoblastic leukemia (ALL). After isolation of genomic DNA we selectively amplified codons 12,13 or 60,61 of the RAS genes and the FLR exon of NF1 by polymerase chain reaction (PCR). Afterwards, all PCR products were directly sequenced by the chain termination method. We found only one point mutation in codon 13 of the K-RAS gene (GGC to GAC) leading to an amino acid exchange from glycine to aspartic acid. All other RAS genes as well as the FLR exon of NF1 were found to be of wild-type DNA sequence. We conclude that the RAS protooncogenes and the NF1 gene do not play a major role in the development of secondary leukemias.
