Zusammenfassung
Die Bedeutung der erhöhten Produktion von Stickstoffmonoxid (NO) bei chronisch entzündlichen Gelenkerkrankungen, aseptischen Lockerungen von Endoprothesen sowie der Heilung von Bandstrukturen ist ungeklärt. NO wird im Rahmen dieser Prozesse durch die in Chondrozyten, synovialen Fibroblasten und aktivierten Leukozyten exprimierte induzierbare NO-Synthase (iNOS) freigesetzt [1]. Tierexperimentelle Daten deuten auf positive Effekte einer Blockade der iNOS auf den Krankheitsverlauf der rheumatoiden Arthritis hin [2]. Andererseits sind anti-adhäsive Eigenschaften von NO auf die Leukozyten-Endothelzell-Interaktion nachgewiesen worden [3]. Das Ziel unserer Studie war daher, die Bedeutung der iNOS-Expression für die synoviale Mikrozirkulation von Mäusen mit Antigen- induzierter Arthritis (AiA) zu untersuchen. Hierfür wurde die in unserem Labor entwickelte Methode zur intravitalmikroskopischen Analyse der synovialen Mikrozirkulation im Maus-Kniegelenk angewendet [4].
