Abstract

This chapter describes the testing of iodinated angio- and urographic x-ray contrast media before clinical testing. Once a promising series of chemical compounds has been conceived, a range of physicochemical and toxicological tests will be used to select the most promising candidate(s), which then will be subjected to a wider range of tests, including stability, pharmacokinetic, and toxicological tests, in order both to make the final choice and to predict its expected safety in man before clinical testing. After a new compound has been synthesized, and has been shown to be chemically stable, the major factor determining its suitability as a contrast agent is its toxicology. Much of this chapter is therefore devoted to aspects of toxicological testing.
