Summary
The expression of genes of the MAGE family which encode tumor-rejection antigens recognized by autologous cytotoxic T-lymphocytes in association with a major histocompatibility complex class I molecule, has potential for the development of the specific immunotherapy of HCC. Methods: We studied 13 HCC specimens using the reserve transcription-polymerase chain reaction (RT-PCR). Results: One or more genes were expressed in 12 of 13 cases (92.3%). Six samples expressed between two and five MAGE-genes. The expression of melanoma antigen encoding-genes 1, 2, 4, 6, 9, 10 and 12 was observed in 23%, 15.4%, 53.8%, 15.4%, 7.7%, 7.7% and 53.8%, respectively. Of the 13 patients, five (38.5%) had a hepatitis-associated HCC. Conclusions: MAGE-gene-products are expressed in a high frequency and suggest the possibility as a target for tumor-specific immunotherapy for HCC-patients.
Zusammenfassung
Fragestellung: Die Gene der MAGE-Familie verschlüsseln tumorspezifische Antigene. Die Peptide werden zytotoxischen T-Lymphozyten über bestimmte MHC-Klasse-I-Moleküle präsentiert und stellen eine potentielle Grundlage für eine spezifische Immuntherapie dar. Material und Methodik: Aus kryoasservierten Gewebeproben des HCC wurde Total-RNA isoliert. Anschließend erfolgte die Synthetisierung der cDNA und die Amplifizierung mit der RT-PCR. Ergebnisse: Eine Expression von einem oder mehreren MAGE-Genen wurde bei 12 von 13 Tumoren (92,3%) beobachtet. 6 Tumore exprimierten zwischen 2 und 5 MAGE-Genen. Die Expression der Peptide 1, 2, 4, 6,9,10 und 12 erfolgte zu 23%,15,4%, 53,8%,15,4%, 7,7%,7,7% und 53,8%. Bei 5 von 13 Patienten wurde eine Hepatitis-Infektion nachgewiesen. Schlußfolgerung: Aufgrund der hohen Expressionsrate der MAGE-Gene beim HCC könnte sich bei dieser Tumorentität die Möglichkeit für eine adjuvante, spezifische, antitumorale Immuntherapie eröffnen.
