Pharmakodynamik und Pharmakokinetik beim Intensivpatienten, Interaktionen

Zusammenfassung

Die Pharmakodynamik beschreibt die Einflüsse eines Pharmakons auf den Organismus, befasst sich also mit der Art und Weise seiner biologischen Wirkungen. Sie beinhaltet Überlegungen zu Wirkungsmechanismen, Rezeptortheorien, Dosis-Wirkungs-Beziehungen und Nebenwirkungen.

Pharmaka sind in der Lage, sowohl spezifische Wirkungen, die über Rezeptoren vermittelt werden, als auch unspezifische Wirkungen, die meist von den physikalischen Eigenschaften der Substanz abhängig und nicht rezeptorabhängig sind, zu vermitteln. Zur Auslösung spezifischer Wirkungen sind, anders als bei den unspezifischen Wirkungen, nur relativ niedrige Dosen bzw. Konzentrationen des Pharmakons notwendig, und es gibt einen genau definierbaren Angriffsort. Sowohl die spezifische als auch die unspezifische Bindung eines Wirkstoffes führt zu charakteristischen Veränderungen auf subzellulärer und molekularer Ebene, die zu wenigen grundlegenden Reaktionsmustern zusammengefasst werden können.
