Stratum Corneum Lipids and Filaggrin

Abstract

Stratum corneum, the most upper layer of the skin, plays a key role for the skin barrier function. The stratum corneum mainly consists of corneocytes and lipids. The lipids are divided into ceramides, cholesterol, and free fatty acids. The synthesis of the lipids is regulated by enzymes, and the enzymes are influenced by pH due to the various pH optima of the different enzymes involved.

As filaggrin breakdown products contribute to the acidic pH of the stratum corneum, there is a possibility that filaggrin deficiency could influence stratum corneum lipids. However, despite people with a filaggrin gene mutation having a more neutral pH, the influence on the lipids is likely to be minimal.
