Abstract
Severe demands and limitations are imposed on any intracellular analytical technique by the environment within the living cell. These difficulties arise both from the nature of the biological system and from the technical challenge of the intracellular approach. The cellular fluid consists of a highly complex sample of extremely small volume and is surrounded by a membrane which is crucially involved in the preservation of the state and function of the cell. In order to investigate this space in a single cell a highly selective and extremely miniaturized technique is required.
