Zusammenfassung
Die Isolierung und quantitative Bestimmung von Proteinen, Aminosäuren und Aminen setzt in der Regel eine wäßrige Extraktion aus dem betreffenden biologischen Material voraus, sei es tierischer, pflanzlicher oder bakterieller Art. Nur bei Körperflüssigkeiten wie z. B. dem Blutserum, Drüsensekreten (Milch) liegt das Protein schon in freier Form in Lösung vor. Vor allem, wenn es sich um tierisches Gewebe handelt, ist die Methode der Wahl eine grobe mechanische Zerkleinerung mit einem Fleischwolf, dem eine Homogenisierung mit einem Starmix, in der amerikanischen Literatur auch „Waring Blendor“ genannt, folgt. Unter diesen Bedingungen wird meist unter Zusatz von Wasser oder Pufferlösung ein Extrakt erhalten, der bei möglichst hochprozentiger Zerstörung der Zellen und nach Wiederholung der Extraktion mit frischem Lösungsmittel nahezu das gesamte lösliche Protein enthält. Bei Alkohol-löslichen Proteinen müssen natürlich besondere Extraktionsverfahren angewandt werden. Hier sei als Beispiel aufgeführt die Extraktion von Gliadin aus Weizenmehl nach D. B. Dill u. Mitarb. (1925).
