Zusammenfassung
Das Binnenskelett aus elastischem Knorpel, harten Knochen, festen Sehnen und Bändern ermöglicht Wirbeltieren und dem Menschen eine Existenz und Größenentwicklung außerhalb des Wassers. Es funktioniert als z. B. Stabilisator der labilen Organsysteme. Die Knorpelteile des Skeletts bestehen aus elastischen Mukopolysacchariden und faserigen Bestandteilen, den Kollagenfasern, die von Knorpelzellen (Chondrocyten) in den Interzellularraum abgegeben werden. Die Kollagenfasern sind unterschiedlich stark durch kovalente Bindungen miteinander verbunden, sehr stark z. B. in Bändern und Sehnen. Die Knochen bestehen ebenfalls aus einer von Osteoblasten gebildeten Matrix, die neben Kollagen Mineralien vorwiegend Calciumphosphatverbindungen enthält. Die Calciumeinlagerung wie auch der Calciumverlust der Knochen ist ein überaus komplex geregelter Prozess, an dem u. a. das Parathormon und Vitamin D (Calcitriol) wesentlich beteiligt sind. Im Alter nehmen die Knorpelsubstanz und die gesamte Knochenmasse ab (Osteoporose), was ein erhöhtes Bruchrisiko zur Folge hat. Osteoporose ist eine der häufigsten Alterskrankheiten, die vorbeugend und akut durch Ernährung mit Calcium- bzw. Vitamin- D-haltigen Nahrungsmitteln (Fischöle) behandelt werden kann. Ein molekularer Mechanismus der Alterung von Knorpel besteht in der Bildung von Glucose- und Pentosebrücken zwischen den Kollagenfasern, die zu einer Verringerung der Elastizität führen. Die Demineralisierung des Knochens hängt eng mit dem Calciumgehalt des Blutes und der Calciumresorption im Darm zusammen, die im Alter vermindert ist. Auch neuronale Veränderungen der Aktivität des sympathischen Nervensystems sowie Entzündungscytokine haben einen wesentlichen Einfluss darauf.
