Cytomegalovirus membrane envelopment revisited — a STEM-tomography study based on high-pressure freezing and freeze substitution

Abstract

Membrane envelopment of viruses, such as secondary envelopment of human cytomegalovirus (HCMV) has been investigated in the electron microscope since many years. In this study we have investigated this process with novel electron microscopical methods like high-pressure freezing (which allows for arrest of dynamic events) freezesubstitution and scanning transmission electron microscopical (STEM) tomography [1] (which enables three-dimensional data collection of relatively thick sections up to 500 nm). The method is ideal to investigate membrane envelopment of viruses, since this event is fast and an enveloped cytomegalovirus (150 nm) fits well into these sections.
