Die Organellbezeichnung „Peroxisom“ wurde 1966 von de Duve und Baudhuin eingeführt und geht auf den Gehalt an Wasserstoffperoxiden zurück, die durch Oxidasen gebildet und durch Katalase abgebaut werden. Peroxisomen kommen in allen menschlichen Zellen mit Ausnahme der Erythrozyten vor. Die Organellmembran besteht aus peroxisomenspezifischen integralen Membranproteinen mit meist noch unbekannter Funktion. Die peroxisomale Matrix enthält mehr als 50 Enzyme für eine Vielzahl anabolischer Funktionen, wie z. B. Plasmalogenbiosynthese, Cholesterol- und Gallensäuresynthese, und auch katabolischer Stoffwechselwege, wie z. B. Abbau von Wasserstoffperoxiden, von überlangkettigen Fettsäuren (VLCFA) und von Phytansäure.
