Zusammenfassung
Im Serum von 84 Neugeborenen und Erwachsenen (0–101 Jahre) wurde die Kinetik der Bindung an Humanserumalbumin in Abhängigkeit vom Lebensalter untersucht. Dansylsarkosin (DS) assoziiert spezifisch an der Benzodiazepinbindungsseite von Humanserumalbumin und dient als Modelligand. Die Bestimmung der Bindungskinetik erfolgte in einer Durrum-Gibson Stopped-flow Apparatur über die Aufzeichnung des Zeitverlaufs der Fluoreszenz von Dansylsarkosin während der Bindungsreaktion. Mit einer rechnerischen Anpassung wurden Exponentialfunktionen und Relaxationskonstanten (krel) aus den Fluoreszenzzeitverläufen ermittelt. Aus diesen konnten die Geschwindigkeitskonstanten für die Assoziation (k2) und Dissoziation (k−2) sowie die Affinitatskonstanten (KA, K ′A ) für die Bindung von Dansylsarkosin an Humanserumalbumin berechnet werden.
Ein signifikanter linearer Zusammenhang besteht zwischen dem Lebensalter und der Assoziationsgeschwindigkeitskonstanten k2 (r2 = 0,59; P < 0,001). Im Serum von Neugeborenen (Mittelwert: k2 = 300 s−1, n = 32) ist k2 größer als im Serum von Erwachsenen im Alter von 23∓65 Jahren (k2 = 200 s−1; n = 28, P < 0,001) und von Erwachsenen im Alter von 66∓101 Jahren (k2 = 144 s−1; n = 24, P < 0,001), so daß die Assoziationsgeschwindigkeit in dieser Reihenfolge abnimmt. Keine wesentliche Altersabhängigkeit besteht für die Geschwindigkeitskonstante k−2 (= 18 s−1). Die Affinitätskonstanten KA sind daher im Alter geringer (Neugeborene: KA = 4,90 105 l/mol;23∓65 Jahre: 4,84∓105 1/mol, n.s.; 66∓101 Jahre: 2,23 105 1/mol, P < 0,001).
In Seren mit einem geringen Wert von k2 wurde eine erhöhte Konzentration an freien Fettsauren gemessen: es besteht ein deutlicher Zusammenhang zwischen der Abnahme von k2 und der Zunahme der Konzentration freier Fettsäuren bei alten Menschen. Das molare Konzentrationsverhaltnis aus freien Fettsauren und Humanserumalbumin beträgt im Mittel 0,64 bei Neugeborenen, 0,61 bei Erwachsenen (23∓65 Jahre) und 1,40 im Alter (66∓101 Jahre, P < 0,001).
In einem synthetischen System ausgereinigtem Humanserumalbumin, Dansylsarkosin und Oleat wurde mit fast identischem Ergebnis bestätigt, daß an das Albumin gebundene freie Fettsäuren die Dansylsarkosin-Bindung hemmen. Freie Fettsäuren scheinen somit über einen allosterischen Mechanismus die Konformation an der Benzodiazepinbindungsstelle so zu beeinflussen, daß die Bindungskonstante dort zurückgeht und der ungebundene Anteil des Liganden steigt.
Summary
Human serum albumin is able to specifically bind many endogenous and exogenous substances in blood. The binding kinetics of albumin as a function of age were investigated in serum of 84 subjects (0 to 101 years). Dansylsarcosine (DS) which associates specifically at the benzodiazepine binding site was used as a marker ligand. Binding kinetics were measured in a stopped flow apparatus (Durrum-Gibson) by detection of DS-fluorescence development during the binding reaction. Exponential functions and relaxation constants describing the fluorescence development curves were obtained by curve fitting. Velocity constants for association (k2) and dissociation (k−2) and binding constants (KA, K ′A ) were calculated.
A significant linear relationship (r2 = 0,59, P < 0.001) between velocity constant and age of individuals was obtained. Association is faster in serum of newborn infants (mean: k2 = 300 s−1, n = 32) than in serum from adults (age 23–65 years, k2 = 200 s−1, n = 28, P < 0.001) and is slowest in serum of elderly subjects (66–101 years, k2 = 144 s−1, n = 24, P < 0.001). The rate of dissociation (k−2 = 18 s−1) is not age dependent and so the binding constant KA decreases with age (newborns: 4.90 105 l/mol; 23∓65 years: 4.84 105 1/mol, ns; 66∓101 years: 2.23 105 1/mol, P < 0.001).
There is a close relationship between k2 and the concentration of free fatty acids in serum, which increases with age. In newborns and adults (23–65 years) the mean free fatty acid/albumin molar ratios were 0.64 and 0.61 whereas in the elderly the molar ratio was 1.4 (P < 0.001).
The role of free fatty acids was confirmed by measurement of the binding kinetics of DS to purified albumin in a synthetic system containing oleate. This showed that free fatty acids apparently have the potential to induce conformational changes at the benzodiazepine binding site of human serum albumin thereby lowering the binding constant and increasing the unbound fraction of drug.
