Complement Fixation Test (CFT)

Abstract

Complement fixation is a process that initiates through the binding of C1 to at least two IgGs or one IgM, triggering the classical pathway to form a membrane attack complex (MAC), and eventually the lysis of the target. This reaction is utilized in clinical testing because complement can lyse any antibody–antigen complexes. Here, the principle, reagents required in addition to the preparation steps and results interpretation for the complement fixation test are explored.
