Molecular Biology of JC Virus and the Human Demyelinating Disease, Progressive Multifocal Leukoencephalopathy

Abstract

JC virus (JCV) is a human neurotropic polyomavirus that causes the fatal demyelinating disease of the central nervous system known as progressive multifocal leukoencephalopathy (PML). JCV was first isolated in 1971 from the brain tissue of a patient suffering from PML. With the emergence of AIDS in the 1980s, the incidence of PML increased dramatically. There has been a rapid expansion in the number of the additional human polyomaviruses in recent years, due to new discoveries, bringing the present number to 13. In his chapter, a particular attention was devoted to the description of the molecular biology of JCV (section “The Molecular Biology of JCV”) and the pathophysiology of PML (section “The Pathology and Pathogenesis of Progressive Multifocal Leukoencephalopathy”) with special emphasis on exciting aspects of current research into this increasingly important pathogen.
