Abstract

Vascular Endothelial Growth Factor A (VEGF-A) is a secreted growth and survival factor that binds to endothelial cells and plays a critical role in the induction of angiogenic processes. It is essential for embryonic development and during adulthood plays a key role in physiological processes that require angiogenesis, such as the growth and differentiation of tissues, wound healing, the menstrual cycle and placenta formation. VEGF-A also plays an important role in the progression of cancer and other diseases which require the formation of new blood vessels, such as proliferative retinopathies, arthritis and psoriasis. Thus, its expression must be tightly controlled since dysregulation has disastrous consequences. The regulation of VEGF-A expression is an incredibly complex, convoluted and wide-ranging exhibition of the scope of eukaryotic post-transcriptional regulation. This chapter will describe our current knowledge of the many post-transcriptional regulatory mechanisms that are used to modulate cellular VEGF-A levels, and will show why this gene is commonly used as a paradigm for post-transcriptional regulation.
