Abstract
In a tissue, the protein expression may differ depending on the position in the tissue. A common proteomic approach, analyzing the protein expression of homogenized tissues, would not represent such localized changes of expression. Then, in order to measure the protein expression within the tissues, in late 1990’s a method of mass spectrometry analyzing intact and fresh tissues has been developed. This method, called mass spectrometry imaging, started using MALDI ion source and has now been expended to ESI ion source with for example DESI and LAESI techniques. On the other hand, the direct analyses of biological samples, such as biopsy or blood sample, are being developed in order to utilize mass spectrometry as a diagnostic tool. This chapter presents some of the mass spectrometry imaging and diagnostic methods and their development.
