Protein Peroxides: Formation by Superoxide-Generating Systems and During Oxidation of Low Density Lipoprotein

Summary

BSA incubated in solution in presence of several superoxide producing systems acquired peroxide groups which were detected by an iodometric assay. Activated neutrophils, xanthine oxidase acting on xanthine in presence of Fe (III), NADH oxidase with NADH and Fe (III) or Cu (II), and the combination of ascorbate/Fe(II)/EDTA were all effective. The role of Fenton reaction in the oxidation was shown by its inhibition with SOD, catalase, or absence of metal. Apoprotein B isolated from human LDL oxidised by gamma radiation or by incubation with Cu (II) was also peroxidised. The extent of peroxidation was linear with time, showing that the process was not affected by the normal antioxidants present in the LDL particles.
