Summary

Chronic application of nicotine to rats and guinea pigs for 8 weeks induces species dependent changes in blood platelet and cardiac function, which can be ascribed to elevated levels of noradrenaline in plasma and tissue. Platelet aggregability in the rat is attenuated by a β-adrenoceptor mediated action on adenylyl cyclase, there being no effect on the adrenoceptor-devoid platelets of the guinea pig. The β-adrenoceptor density of rat myocardium is down-regulated in 1 week upon nicotine application, affording protection of the heart from any untoward metabolic stress due to elevation of catecholamine release. In contrast, ß-receptor down-regulation occurs more slowly in the guinea pig myocardium (4 weeks). A transitory depression of function can be observed in working heart preparations isolated after 2 weeks of pretreatment. This is associated with lower levels of ATP and NAD+ in the ventricular tissue. The metabolic signs of stress disappear again with progression of down-regulation.
