Abstract
The successful preparation of nuclear extracts capable of performing in vitro transcription depends on careful manipulation at each step of the protocol. The development of such a method, on the other hand, depends mainly on the careful assessment of theoretical as well as practical steps likely to influence the quality of the end product.
