Abstract
This chapter deals in depth with the topics of spillover and compensation. As for the spillover, the chapter provides an exhaustive description of the phenomenon and its occurrence in the context of multicolor cytometric analysis, distinguishing the various possible types and relating them to the spectral characteristics of the fluorochromes. The chapter then describes the spillover and compensation matrices and considers their role in the compensation process, paying particular attention to the different circuital architectures. Finally, the chapter considers the paradoxical effects that appear once the compensation is reached, including the production of negative values, the spreading spillover, and the perturbation of the distribution of the negative component.
