Detection of hrHPV DNA with Simulated HPV16 and HPV18 Typing Based on Real-Time PCR

Abstract

The complexity of molecular diagnosis of HPV that is directly related to cervical cancer is caused by a large number of genotypes, such as HPV genotypes 16 and 18, which show the greatest malignant potential and cause 70% of all cervical cancers. The objectives of this study are to determine the significance of Ct values detected by HPV DNA as well as intern control DNA in cervical smear samples and to evaluate the advantages and disadvantages of cervical cancer screening based on HPV DNA and to determine the incidence of HPV16, HPV18 and other mixed infections. In this report, the Abbott RealTime High-Risk HPV Diagnostic Test (Abbott Molecular Inc., IL, USA), for the presence of hrHPV infection, routinely tested a total of 1419 cervical samples of women with cytological changes on the cervix. The average age was 42.59 years. The prevalence of hrHPV infection was 29.81% (hrHPV16, 18 and others). Ct HPV18 showed a significant difference between age groups. Approximately constant values of Ct-IC samples of cervical swabs, Ct values of viral DNA could be considered as a parameter of disease progression/regression. A statistically significant correlation was found between Ct-IC and Ct-HPV16.
