Regulation of Cell-Matrix Adhesion Networks: Insights from Proteomics

Abstract

Cell adhesion to the extracellular matrix regulates many fundamental aspects of cellular behaviour, including cell survival, proliferation, differentiation, and migration. Cell-matrix interactions are mediated by integrin-associated adhesion complexes, multiprotein complexes of intracellular scaffolding and signalling proteins that link the cytoskeleton to the extracellular milieu. These linkages enable cells to sense and respond to the biochemical and biomechanical properties of the microenvironment. Advances in proteomic methodologies have enabled the characterisation of adhesion complex composition, interactions, and dynamics, which has led to new insights into the physical and functional properties of cell-matrix adhesion networks. These approaches have broadened our understanding of the molecules that regulate cell adhesion to the extracellular matrix.
