Abstract
Human embryonic stem cells (hESCs) are pluripotent cells derived from the inner cell mass (ICM) of the developing embryo. They have tremendous potential for the research of early human development, differentiation processes, and teratology, as well as for industrial and clinical purposes, such as drug screening and cell-based therapy. Since hESCs were first derived by Thomson and his colleagues in 1998, considerable effort has been invested to improve methods of isolating new lines, in defined conditions, and to increase success rates. This chapter discusses the most commonly used methods for deriving hESC lines, including immunosurgery and the whole embryo approach.
