Abstract

Apoptosis is a form of cell death that is mediated by specific biochemical cascades involving mitochondrial changes and activation of proteases called caspases. Apoptosis provides a mechanism for cells to die without adversely affecting their neighbors (Fig. 1). This contrasts with a nonregulated form of cell death called necrosis, in which cellular organelles swell and the plasma membrane becomes permeable, resulting in release of cellular contents and massive death of groups of cells throughout a tissue. Aberrant regulation of apoptosis may be the central abnormality in many different diseases, most prominent of which are cancers wherein cells become resistant to apoptosis. Therefore, a major effort in the field of cancer research is to identify the molecular alterations responsible for such cell immortalization (Wyllie, 1997).
