Abstract

Mutant analysis is a powerful tool in the study of biological processes, the more so if it is combined with the relevant physiological experiments. Two basically different approaches can be used for the construction of mutants: (i) mutants can be obtained with chemical mutagens or by irradiation with UV or X-rays; (ii) mutants can be generated by insertional mutagenesis. The basic idea behind the latter method is the insertion of an easily selectable genetic element into the gene of interest. This has the major advantage that the gene is marked in such a way that it can be isolated easily.
