Platelet-Activating Factor (PAF): Signalling and Adhesion in Cell-Cell Interactions

  • Chapter
Platelet-Activating Factor and Related Lipid Mediators 2

Abstract

PAF is a signalling molecule1 that mediates interactions between cells2. Cell-cell interactions are critical in both physiologic and pathologic processes3,4. Many of the best-characterized effects of PAF involve cellular signalling in the inflammatory and vascular systems, but it also accomplishes signalling functions in the reproductive system, the brain, and in many other organs2. It has the remarkable property of being able to act as an endocrine, paracrine, autocrine, or juxtacrine signal depending on the cell of origin and the specific conditions under which it interacts with target cells5. The amphipathic properties of PAF and its ability to interact with both lipids and proteins contribute to this versatility.

Supported by the Nora Eccles Treadwell Foundation, individual grants from the National Institutes of Health and the Department of Veterans Affairs Medical Research Fund, an NIH Training Grant in Cellular and Physiologic Responses in Lung Disease, an NIH Special Center of Research in ARDS, and an NIH Center of Excellence in Molecular Hematology.
