Response of Brain Endothelial Cells and Bone-Marrow Macrophages to Leukocyte-Derived Effectors

Summary

Activation of endothelial cells is involved in the recruitment of blood-borne cells into organs. In the cerebral vascular system, perivascular macrophages of bone marrow origin are found directly underneath the endothelial layer and are postulated to participate in endothelial activation. To evaluate the respective role of endothelial cells and bone marrow-derived macrophages, we compared their responses to inflammatory stimulation. Our results suggest that both endothelial cells and macrophages contribute in mediating inflammatory activation of the blood-brain barrier, but also indicate specific modes of regulation for some functions.

Résumé

Une activation des cellules endothéliales est nécessaire au recrutement dans les organes de cellules circulantes. Au niveau du système vasculaire cérébral, des macrophages périvasculaires originaires de la moelle osseuse sont localisés dircetement sous la couche endothéliale et participent vraissemblablement à l’activation endothéliale. Dans le but d’évaluer le rôle des cellules endothéliales et des macrophages dans le processu d’activation, nous avons comparé leurs réponses à une stimulation inflammatoire. Nos résultats suggèrent que les deux types de cellules participent au processus inflammatoire au niveau de la barrière hémato-encéphalique, mais que les phénomènes régulatoires sont spécifiques.
