Urinanalysis by 1H NMR: Application to Cephaloridine-Treated Rats
Although a quite recent development the analysis of biological fluids by high-resolution proton NMR is rapidly becoming an established procedure, especially for urine and plasma (e.g. [1–3], and arts, by J.K. Nicholson & colleagues in this book and in Vols. 16 & 17). A particular advantage is the wealth of additional information furnished, over and above that required for the qualitative or quantitative determination of a single analyte that is manifest in the spectrum from the sample.- Any organic molecule possessing protons, will be detected, provided that the levels are mM or higher. Accordingly, a single assay, on otherwise untreated urine samples, can comprehend drugs, drug metabolites, and endogenous metabolites (e.g. Krebs cycle intermediates, sugars, organic acids, creatinine and various amino acids) that may bear on drug-related toxicicity. No Chromatographic method could be as simple and wide-ranging as 1H NMR. These points are now exemplified by 1H NMR observations on urine of rats dosed with a nephrotoxic cephalosporin antibiotic, cephaloridine.
