Nifedipine (NF), nitrendipine (NT) and nimodipine (NM) are potent calcium-entry antagonists that modify calcium-channel activity in different smooth muscles. As therapeutic agents they are widely used in the treatment of coronary heart diseases (NF), hypertension (NT) or cerebral disorders (NM). The calcium entry blockade is closely related to the intact dihydropyridine (DHP) ring system. Metabolites are usally pyridine analogues of the parent drugs without any effects on calcium channels; even though they are ineffective and non-toxic, their pharmacokinetic profiles have to be studied.
