Abstract
The present report aims at presenting a synthetic review of the most important clinical trials which use gangliosides as therapeutic agents in peripheral neuropathies of various etiology. The complex of biochemical and pharmacological research studies on gangliosides indicates that these molecules, natural components of cellular membranes (in particular, of neuronal membranes) are involved in neuronal development, differentiation and regeneration processes. The ganglioside preparation has been shown to possess a reinnervation-stimulating activity due to enhanced nerve sprouting, an essential feature of muscular reinnervation processes and of restoration of synaptic contacts. Electrophysiological and functional evidence of early recovery from nerve damage, due to parenteral ganglioside treatment, has been obtained in several animal models, including motor nerve function after nerve crush, sensory nerve function after transection, cochler impairment by noise, diabetic neuropathy in mutant diabetic mice and toxic damage to peripheral nerves (Ceccarelli et al., 1976; Gorio et al., 1980; 1983; 1984; Leon et al., 1981).
