Abstract

Affinity chromatography is now an established technique for the purification of biologically active compounds (1,2). As in all rapidly expanding sciences, new concepts are continually being introduced and old ones modified or discarded. The use of spacer arms to enhance binding was first heralded as a major advance. Gradually it became clear that it could also serve merely to increase nonspecific binding due to the length of the arms, their hydrophobic properties and the charges introduced during the coupling reaction with cyanogen bromide (3).
