Abstract

As part of an enzymological approach to the investigation of proteins involved in the repair of chromosome damage in mammalian cells, we have focused our attention on the enzyme DNA ligase which participates in all the physiological processes involving DNA breakage and rejoining, i.e., DNA repair, recombination and transposition. DNA ligase is also required during DNA replication, since it participates in the replisome complex by catalysing the ligation of Okazaki fragments synthesized in the lagging strand. In all reactions, the enzyme is responsible for covalently joining 3′-hydroxyl to 5′-phosphoryl termini in double-stranded DNA.
