Antibodies have long been envisioned as potential clinical reagents. Although the development of hybridoma technology (Kohler and Milstein, 1975) has unleashed a vast array of possibilities for the generation of monoclonal antibodies (MAbs) for use in diagnosis and therapy of cancer and other diseases, several limitations inherent to MAbs exist. For example, MAbs of the desired specificity may be difficult or even impossible to generate using current hybridoma methodology. Furthermore, the MAb developed may not have the affinity needed for a given function, or the MAb’s isotype may not be optimal for the desired effector function, for example, antibody-dependent cell-mediated cytotoxicity (ADCC).
