The Effect of Acetazolamide and Furosemide on Lithium Clearance and Cisplatin Nephrotoxicity in the Rat

Abstract

The clinical use of cisplatin is limited because of its potential nephrotoxicity. Diuretics such as acetazolamide or furosemide are known to decrease cisplatin nephrotoxicity (Osman et al., 1984; Heidemann et al., 1985). Since mannitol 5% and glucose 30% also have beneficial effects on cisplatin-induced kidney failure (Pera et al., 1979; Heidemann, unpublished data) the question arose whether intratubular dilution of cisplatin in the proximal tubules might contribute the observed protection. It was therefore the aim of this study to compare the lithium clearance during diuretic treatment at the time of cisplatin administration with the kidney function after 5 days.
