Abstract
The previous chapter described the evolution of the receptor concept and the early appreciation for the complexity that can exist between receptor occupancy and the ultimate physiological response. The content of the present chapter (1) summarizes some of the methods used to characterize the specificity of the receptor which elicits an observed response, and (2) outlines methods for determining the affinity constants of agonist, partial agonist, and antagonist agents. It will become clear to the reader that these latter methods exploit early insights regarding efficacy and receptor reserve.
