Preparing Hydra for Scanning Electron Microscopy

Abstract

Scanning electron microscopy (SEM) is used predominantly to study cell surfaces, although slices of tissue or fractured surfaces of fixed or frozen specimens can also reveal some features of internal organelles. Conventional scanning instruments are limited in resolution to approximately 150 Å with most biological material (Everhart and Hayes, 1972). Field emission instruments now coming into more general use are expected to improve this Figure significantly (Crewe, 1971). The scanning electron microscope can also be used for x-ray microanalysis. When used with the scanning transmission mode of operation, important information concerning the presence and distribution of specific elements of biological significance can be obtained (Marshall, 1975).
