Purification and Characterization of Pig Heart AMP-Deaminase
AMP-deaminase (AMP-D) (E.C. 188.8.131.52) catalyzes the hydrolytic deamination of AMP to IMP and NH3. AMP-D plays an important role in skeletal muscle and liver (1,2). We purified AMP-D by adsorption chromatography on cellulose phosphate and ion-exchange chromatography on DEAE-Sephacel. We examined the kinetic constants and the influence of changing energy charges on this enzyme and investigated the difference with the enzyme purified from pig skeletal muscle.
