Renal Peritubular Amyloid Deposition in the Experimental Murine Amyloidosis
In order to define the process of peritubular amyloid formation, kidneys of amyloid laden mice were examined by immunohistochemistry with antimouse AA antibody as well as by conventional light and electron microscopy. The results suggest the following: (a) amyloid precursors reach the basal plasma membrane by two possible routes, ie, from the interstitial capillaries and from the tubular lumen; (b) From the proximal tubules amyloid precursors are reabsorbed by endocytosis and are subject to lysosomal breakdown; (c) a part of the precursors are transported to the basal plasma membrane below which amyloid fibrils are formed; (d) the disappearance of interdigitations and the appearance of deep indentations of the basal plasma membrane are morphologic changes associated with the deposition of amyloid fibrils or their precursor proteins.
