The Interrelation Between CBF, Energy Metabolism and Ecog in a New Awake Brain Model
The measurement of intramitochondrial NADH oxidation-reduction state from the brain of an awake animal is a well established technique and is in use for the last few years. We had used this animal model in studying the effects of hypoxia, spreading depression, epileptiform activity (Mayevsky and Chance, 1975) or ischemia (Mayevsky, 1977) on the metabolic activity. In all of the studies made by our group or other groups, only one site or side of the brain was monitored fluorometrically. In the ischemia studies we found that under certain experimental conditions it is very important to measure metabolic activity from the “experimental” site as compared to the “control” site or side simultaneously. In our awake model we used a topical application technique to study the effects of drugs on the cortical tissue without any effects on the rest of the brain. This model also requires a bilateral measurement technique. We present in this presentation a new type of fluorometer/ reflectometer by which NADH fluorescence, as well as reflected light, can be measured simultaneously from two points of an awake brain model. This model was exposed to anoxia, spreading depression or ischemia and the results are presented. In another group of rats the effects of topical application of 2-Deoxyglucose were tested.
