In Vitro Replication of Bacteriophage Φ29

Abstract

The Bacillus subtilis phage Φ29 has as genetic material a linear, double-stranded DNA of about 18, 000 base pairs (1) with a protein covalently linked to the 5′ ends by a phosphoester bond between serine and 5′ dAMP, the terminal nucleotide at both 5′ ends (2). The protein, product of the viral gene 3, p3 (3), is required for the initiation of replication (4). An inverted terminal repetition, six nucleotides long, exists at the DNA ends (5, 6). Φ29 replication starts at either DNA end and proceeds by strand displacement (7, 8).
