Separation of Ribulose 1,5-Bisphosphate Carboxylase and Oxygenase Activities

Recently it was shown that RuBP carboxylase and RuBP oxygenase activities were separated when the enzymes were prepared at a higher pH than normally used (1). Recent reports (2, 3), as well as some characteristics of the enzyme found earlier (4), e.g., the different behaviors of the two activities upon storage, suggest that two different enzymes are responsible for the two activities. We describe here three different methods for separating the two activities and two spectrophotometry methods for rapidly measuring the RuBP oxygenase activity.
