Abstract
Paracelsus, 16th century Swiss physician and recognized father of Toxicology, is quoted as stating [Klaassen and Doull, 1980] “All substances are poisons; there is none which is not a poison. The right dose differentiates between a poison and a remedy”. Therefore, a functional definition for toxicity would state that virtually every substance possesses the potential to produce injury or death (hazard) if available in a sufficient amount (exposure). This idea encompasses not only the response to the substance but also the delivery of the dose to the target tissue(s), and hence implies hazard and exposure or risk. In the biological model, risk is determined by a dynamic sequence of events; biokinetics, whether restricted to a pharmacological response or an overdose expressing itself as a toxicological response, is central to understanding risk.
