Mechanism of Steroid Hormone Action: In Vitro Control of Gene Expression in Chick Oviduct Chromatin by Purified Steroid Receptor Complexes

Abstract

Numerous studies have demonstrated that steroid hormones appear to exert their primary influence at the level of genetic transcription (O’Malley and Means, 1974). Evidence in endocrine target tissue systems utilizing estrogen (Gorski and Nicolette, 1963; Hamilton et al., 1963), progesterone (O’Malley and McGuire, 1968a,b), aldosterone (Edelman and Fimognari, 1968), glucocorticoids (Kenney and Kull, 1963; Sekeris and Lang, 1964) and vitamin D (Norman, 1966; Stohs et al., 1967) have demonstrated increased RNA synthesis after the in vivo localization of administered hormone in the nuclear compartment. Quantitative and qualitative changes in RNA synthesis usually precede the steroid hormone stimulation of inducible protein biosynthesis in target tissues. Recent experiments, in fact have conclusively demonstrated that female sex steroids are capable of inducing a net increase in specific mRNA molecules in target cells (Chan et al., 1973; Comstock et al., 1972; Harris et al., 1975).
