Use of a Genetic Approach to Study Cyclic AMP Metabolism

In this review, I present a brief overview of a genetic approach for studying cyclic AMP metabolism and the application of this approach with murine S49 lymphoma cells. S49 clones having alterations in the pathway of cyclic AMP generation and function have proved useful in characterizing components involved in hormonal stimulation of adenylate cyclase, in activation of cAMP-dependent protein kinase, and in studying more distal cellular events mediated by cyclic AMP. The use of genetic methods in S49 and other cultured cell systems offers an important complementary approach to biochemical and pharmacological methods for studying the cyclic AMP pathway.
