The Relationship between the Plasma Kinin-System and the Contact Phase of Blood-Coagulation in Man

Abstract

Three mechanisms have been postulated for the activation of plasma kallikrein (Fig. 1). According to the oldest theory activated Hageman factor or Factor Xlla activates the permeability globulin Pf/dil, which then acts on prekallikrein and the latter, as kallikrein, liberates kinin from kininogen (1, 2). The second hypothesis, based on good experimental data, assumes that Factor Xlla acts directly on prekallikrein (3,4,5). More recently, an activator of prekallikrein distinct from Pf/dil has been isolated from guinea pig (6), rabbit (7) and human plasma (8,9,10,11). Some evidence was presented that this prekallikrein activator (PKA) is a fragment of Factor Xlla (8). Accordingly, this substance is designated as Xllf in Fig. 1.
