Antivirals for High Hazard Viruses

In large areas of the world there exist extremely high hazard viruses for which there are no vaccines for prophylaxis and no effective drugs for therapy (Canonico et al., 1984). Examples of such viruses are Ebola, Argentine, Bolivian, Crimean-Congo, and Korean hemorrhagic fevers, Rift Valley fever, Venezuelan equine encephalitis, Japanese B encephalitis and Lassa fever. With the notable exception of the yellow fever vaccine, immunoprophylactic approaches have not been developed for most of these “exotic” viral diseases. Even when effective vaccines are available, vaccination does not provide absolute protection. To compensate for this inadequacy, we recognized the need to develop antiviral drugs which are effective against a broad-range of viruses and which can be used either prophylactically or therapeutically, as required. However, the development of chemotherapy for viral diseases has been slow. The search for substances specifically targeted to viral replication processes has been difficult and shares some parallels with the development of antibiotics.
