Cellulose Bead Entrapped Whole Cell Glucose Isomerase in Fructose Syrup Production
Purified and whole cell glucose isomerase (GI) has been immobilized on various supports (1). Most research has involved Baciteus and Stnep-tomyces enzymes. We have developed a simple and mild technique to entrap whole cell Actinop1anea m-.44ounien4-L4 GI in cellulose (2,3).The method involves suspending dried cells in a-cellulose, dissolved in a melt of N-ethyl-pyridinium chloride (NEPC) and dimethylformamide (DMF). The GI cellulose could be regenerated in fiber or bead form by passing into water through an orifice of dia 0.8 mm, treating with glutaraldehyde for crosslinking, and vacuum drying to about 24% dry weight. This is believed to be the first time an enzyme has been immobilized within regenerated pure cellulose beads.
