Effect of Cold Temperature on Shoot Regeneration in Vitro from Aged Cultures of GF-677 (Prunus Persica × Prunus Amygdalus)
In vitro cultures of GF-677, with arrested growth after a number of subcultures, were placed at 3°C for 4 weeks and then at 22°C. After 4 weeks an average number of 17.6 new shoots per culture were developed, whereas no shoots were formed in cultures which had not received the 3°C temperature treatment. Similar numbers of new shoots were achieved when the cold treatment of 3°C was applied for 3, 4 or 5 weeks, while 1 or 2 weeks in 3°C had no effect on shoot formation. When the cultures of GF-677 with the arrested growth received temperature treatment of 15, 11, 7 or 3°C, a linear response of shoot formation was found, with the minimum number of shoots at 15°C and the maximum one at 3°C.
